During the events of Resident Evil 5, the U.S. space shuttle Columbia is lost above the Atlantic Ocean, and the staff of the Umbrella Corporation return to their corporate headquarters located in Raccoon City. The game begins as Leon S. Kennedy, Jill Valentine, and Claire Redfield arrive at the Umbrella complex, where they are met by the T-Virus and an army of zombies. An Umbrella employee named Ada Wong is still inside the complex, and with her help, the team fight their way to the basement where they find four scientists, who claim to have reverse-engineered the original T-virus to create a cure for the virus.
One year after the events of 4, the then President of the United States, Shepherd Glade has survived an assassination attempt, and the Umbrella Corporation is still under the rule of Raccoon City Police Chief Arthur Birkin. Shortly after this attack, a series of strange murders are committed across the U.S., all involving the use of a child-sized glove. An Umbrella Corporation employee, Jill Valentine, is tasked with investigating the murders, and the FBI is brought in to assist. After witnessing the brutality of the killings, Jill joins forces with Leon S. Kennedy to investigate these crimes. Jill is then assigned to the Umbrella compound in Raccoon City, where she and Leon must fight off the Staff Officer Albert Wesker and his elite soldiers, the S.T.A.R.S., and more.
Bugs were found with, when a character would be downed and the game would begin, they would not be able to move. While this can be fixed with the appropriate controls, it was not fixed on the DS version. When the player falls into a pit or is incapacitated, the game will continue without them. The save feature can be used on a cartridge, but this does not work on all systems. The lack of support for saving on the GBA version of the game was addressed in the 2003 port. Weapon and item names were added to the title screen. The DS version of the game supports the Game Boy Camera for a first-person view.
Residents of the town are scattered among makeshift hideouts, and Leon finds all of them. He meets a plucky survivor named Claire Redfield, who has found a group of other survivors. Leon and Claire are tasked with rescuing the remaining survivors, thus beginning a long hunt for the Umbrella organization.
The game's action, which had been primarily similar to Resident Evil 3, is much more arcade-like. Environments are designed so that combat is more or less unavoidable. On occasion, however, the player may avoid battle entirely by sneaking past enemies.
